Enzymatic and Molecular Identification of Meloidogyne Species in Tomato Orchards in Paraguay

Tomato is a major crop in Paraguay, where it provides source of employment and income for households. production can be affected by root-knot nematodes, especially Meloidogyne spp. The unequivocal identification Paraguay has not been conducted yet. This study aims to identify species eight tomato districts this country biochemical molecular techniques. Females were extracted from roots characterized using esterase isozyme phenotypes. In addition, DNA was nematode eggs, species-specific SCARs (sequence-characterized amplified regions) used confirm the diagnosis. Nematodes detected 100% studied samples (prevalence), which M. incognita (Est: I2, Rm: 1.1;1.2) javanica J3, 1.0, 1.20, 1.35) present 39.13% 26.08% samples, respectively. One population (8.69%) sp. presenting an atypical profile (Rm: 1.0 1.3) only Julián Augusto Saldívar District. Mixed populations, mostly javanica, observed samples. SCAR primers incK14F/incK14R specific fragments (399 bp) (670 bp), confirming enzymatic results. Here, we first at level Paraguay.
